Cells have evolved multiple strategies to adapt the composition and quality of their protein equipment to needs imposed by changes in intra- and extracellular conditions. The appearance of pro teins transmit ting novel functional properties to cells can be controlled at a transcrip tional, posttranscriptional, translational or posttranslational level. Extensive research over the past 15 years has shown that transcriptional regulation is used as the predominant strategy to control the production of new proteins in response to extracellular stimuli. At the level of gene transcription, the initiation ofmRNA synthesis is used most frequently to govern gene expression. The key elements controlling transcription initiation in eukaryotes are activator proteins (transactivators) that bind in a sequence-specific manner to short DNA sequences in the of genes. The activator binding sites are elements of larger proximity control units, ca lied promoters and enhancers, which bind many distinct proteins. These may synergize or negatively cooperate with the activators. The do novo binding of an activator to DNA or, if already bound to DNA, its functional activation is what ultimately turns on a high-level expression of genes. The activity of transactivators is controlled by signalling pathways and, in some cases, transactivators actively partici pate in signal transduction by moving from the cytoplasm into the nuc1eus. In this first volume of Inducible Gene Expression, leading scientists in the field review six eukaryotic transactivators that allow cells to respond to various extracellular stimuli by the expression of new proteins.

This book will deal with heat shock proteins and more generally with stress-related inducible gene expression as a pleiotropic adaptive response to stress. It presents a textbook-like overview of the field not only to heat shock experts, but to physiologists, pharmacologists, physicians, neuropsychologists and others as well. It is intended to be a state-of-the-art and perspective book rather than an up-to-date presentation of recent data. It should provide a basis for new experimental approaches to fields at the edge of the classical heat shock field. Drugs, UV irradiation and environmental toxics will considered as important modulators of the stress response. Radical scavengers such as superoxide dismutases and inducible regulatory proteins of metallic ion status such as ferritin as well as immunophilins and protein disulfide isomerases will be considered within the frame of stress proteins. The potential practical applications of heat shock proteins in toxicology and medicine for the diagnosis, prognosis and eventually therapy of clinical conditions associated with an increased oxidative burden will be outlined. The role of heat shock proteins in the modulation of immune responses will also be included. This book stems from an Advanced Study Institute on Chromo somal Proteins and Gene Expression that was held in Sitges, Spain, on September 17-26, 1984. It would be misleading to call this volume a conference proceedings, however. The ASI was not a conference, but a course with diverse activities, only one of which was a set of major presentations by the lecturers. Indeed, the concept of lecturer was intentionally obscured as we all learned from each other through shorter presentations by other participants and through seminars, poster sessions, and small group discussions. Furthermore, many participants found that exchanging ideas outside organized sessions was among the most rewarding aspects of the course. Some even claimed to have profitably probed the intricacies of nucleosome structure and transcriptional regulation while basking in the sun on the beachl Obviously, it is difficult to catch the flavor of such varied proceedings in a book. (I cannot confirm the incident on the beach, never having found time to set foot there. Such is the fate of the director of a meeting. ) The ASI was judged a success -- and enthusiastically so - by most participants. Not only did we deepen our understanding of our scientific field, we made new friends and learned about scientific and nonscientific aspects of life in other countries and about issues that transcend international boundaries in our complex world. We hope that this volume will be as successful as the course was.
