Spectrally Resolved Detector Arrays for Multiplexed Biomedical Fluorescence Imaging


Book Description

This book describes the design, development, characterisation and application of two novel fluorescence imaging instruments based on spectrally resolved detector arrays (SRDAs). The simplest SRDA is the standard colour camera, which integrates a Bayer filter array of red, green and blue colour filters to replicate the colour sensing capability of the human eye. The SRDAs used in this book contain many more colours, ranging from 16 to over 100 colour channels. Using these compact, robust and low-cost detectors for biomedical applications opens new avenues of exploration that were not possible before, in particular, the use of spectral imaging in endoscopy. The work presented shows for the first time that not only can this new type of camera be used for fluorescence imaging, but also that it is able to resolve signals from up to 7 different dyes – a level of multiplexing not previously achieved in tissue with such compact and robust equipment. Furthermore, it reports the application of a bimodal endoscope performing both reflectance and fluorescence imaging using these cameras in an ex vivo pig oesophagus model.

This book includes some very recent applications and the newest emerging trends of hyper-spectral imaging (HSI). HSI is a very recent and strange beast, a sort of a melting pot of previous techniques and scientific interests, merging and concentrating the efforts of physicists, chemists, botanists, biologists, and physicians, to mention just a few, as well as experts in data crunching and statistical elaboration. For almost a century, scientific observation, from looking to planets and stars down to our own cells and below, could be divided into two main categories: analyzing objects on the basis of their physical dimension (recording size, position, weight, etc. and their variations) or on how the object emits, reflects, or absorbs part of the electromagnetic spectrum, i.e., spectroscopy. While the two aspects have been obviously entangled, instruments and skills have always been clearly distinct from each other. With HSI now available, this is no longer the case. This instrument can return specimen dimensionalities and spectroscopic properties to any single pixel of your specimen, in a single set of data. HSI modality is ubiquitous and scale-invariant enough to be used to mark terrestrial resources on the basis of a land map obtained from satellite observation (actually, the oldest application of this type) or to understand if the cell you are looking at is cancerous or perfectly healthy. For all these reasons, HSI represents one of the most exciting methodologies of the new millennium.

This volume focuses on Time-Correlated Single Photon Counting (TCSPC), a powerful tool allowing luminescence lifetime measurements to be made with high temporal resolution, even on single molecules. Combining spectrum and lifetime provides a “fingerprint” for identifying such molecules in the presence of a background. Used together with confocal detection, this permits single-molecule spectroscopy and microscopy in addition to ensemble measurements, opening up an enormous range of hot life science applications such as fluorescence lifetime imaging (FLIM) and measurement of Förster Resonant Energy Transfer (FRET) for the investigation of protein folding and interaction.
